Acalabrutinib 100 MG Capsules, a highly selective, potent, Bruton tyrosine kinase (BTK) inhibitor is used for the treatment of patients with mantle cell lymphoma (MCL) (who have previously been treated with at least 1 therapy), and chronic lymphocytic leukemia (CLL) or small lymphocytic lymphoma (SLL).

Dosage and Side Effects

The recommended dosage of acalabrutinib capsules as monotherapy for patients with MCL, CLL/ SLL is 100 mg administered orally with or without food approximately every 12 hours until disease progression or if unacceptable toxicity occurs. For patients with previously untreated CLL/SLL, the recommended dosage of Acalabrutinib in combination with the medicine obinutuzumab is 100 mg administered orally with or without food approximately every 12 hours until disease progression or in case of unacceptable toxicity. Swallow capsules whole with water without chewing, or crushing them.

In case a dose of Acalabrutinib is missed by an excess of 3 hours, skip it and take the very next dose at its regularly scheduled time. Additional capsules should not be taken in order to make up for a missed dose.

The most common side effects of Acalabrutinib are upper respiratory tract infection, musculoskeletal pain, neutropenia, anemia, thrombocytopenia, headache, and diarrhea.

Warning and Precautions

  • Critical and serious infections, including opportunistic infections, may occur in individuals with hematologic malignancies treated with Acalabrutinib. Consider prophylaxis in patients who are at higher risk for opportunistic infections. Monitor patients for signs and symptoms of infection and treat them promptly.
  • Fatal and severe hemorrhagic events may occur in those with hematologic malignancies treated with Acalabrutinib. Use of antithrombotic agents concomitantly with Acalabrutinib may further increase the probability of hemorrhage. Monitor patients for probable signs of bleeding.
  • Grade 3/4 cytopenias, including anemia, neutropenia, thrombocytopenia, and lymphopenia, may develop in those with hematologic malignancies treated with Acabrunat 100 mg. Monitor complete blood counts (CBC) regularly during therapy. Interrupt therapy, scale down the dose, or discontinue therapy as warranted.
  • Second primary malignancies (SPM), including skin cancers and certain other solid tumors, may be established in those exposed to Acalabrutinib. Skin cancer is the most frequently occurring second primary malignancy. Monitor patients for signs and symptoms of skin cancers and advise protection from sun exposure.
  • Grade 3 atrial fibrillation or flutter may develop in patients treated with Acabrunat capsules. The risk may be higher in patients with cardiac risk factors, previous arrhythmias, hypertension, and acute infection. Monitor for symptoms of arrhythmia (such as dizziness, palpitations, syncope, or dyspnea) and manage as appropriate.
