Daclatasvir tablet, supplied under the generic brand Natdac, is a hepatitis C virus NS5A inhibitor approved for use with the drug sofosbuvir, either with or without ribavirin, for the treatment of patients with chronic hepatitis C virus (HCV) genotype 1/3 infection.

Dosage and Side Effects

The recommended dosage of Daclatasvir tablets is 60 mg, taken orally, once daily, with or without food along with sofosbuvir with or without medicine ribavirin. The dosage can be reduced to 30 mg once daily with strong CYP3A inhibitors and can be increased to 90 mg once daily with moderate CYP3A inducers. Continue the treatment for 12 weeks. 

The most common side effects of Daclatasvir along with sofosbuvir are headache and fatigue, and along with medicines sofosbuvir and ribavirin are fatigue, headache, nausea, and anemia.

Warning and Precautions

  • Cases of symptomatic bradycardia and cases sometimes requiring pacemaker intervention may develop when amiodarone is coadministered with sofosbuvir together with another HCV direct-acting antiviral medicine, including Daclatasvir. Coadministration of medicine amiodarone with Daclatasvir tablets along with sofosbuvir needs to be avoided.
  • Reactivation of hepatitis B virus (HBV) may develop in those coinfected with HCV/HBV who are undergoing or completed treatment with HCV direct-acting antiviral medicines, and who are not taking HBV antiviral drugs. Test all patients for the existence of current or prior HBV infection by assessing HBsAg or/and antiHBc before starting HCV treatment with Daclatasvir 30 mg/60 mg.
  • Those with signs or symptoms of bradycardia must seek prompt medical evaluation. Signs and symptoms of bradycardia may include chest pain, malaise, near-fainting or fainting, dizziness or lightheadedness, shortness of breath, weakness, confusion, extreme tiredness, or memory problems.
  • In case sofosbuvir is permanently stopped in those receiving Daclatasvir therapy with sofosbuvir, then treatment with Daclatasvir 30 mg/60 mg should also be stopped or discontinued.  
  • Screen patients for the presence of NS5A polymorphisms at amino acid positions M28, Q30, L31, and Y93 in individuals with cirrhosis who are infected with hepatitis C virus genotype 1a before starting therapy with Daclatasvir and sofosbuvir with or without ribavirin.
