Ceritinib Capsule is supplied under the generic brand Noxalk and in the form of oral capsules. Ceritinib 150 mg Capsules is a kinase inhibitor used for the treatment of adult patients with metastatic non-small cell lung cancer (NSCLC) whose tumors are anaplastic lymphoma kinase (ALK)-positive.

Dosage and Side Effects

The recommended dosage of Ceritinib capsule is 450 mg orally (by mouth) once daily with food until the disease progresses or unacceptable toxicity occurs. If a dose is missed, make up that dose unless the next dose is due within at least 12 hours. If vomiting occurs during the course of therapy, do not take an additional dose and continue with the very next scheduled dose of Ceritinib.

The most common side effects of Ceritinib 450 mg with food are abdominal pain, diarrhea, nausea, vomiting, and fatigue; and with Ceritinib 750 mg under fasted conditions: diarrhea, nausea, vomiting, fatigue, weight loss, abdominal pain, and decreased appetite.

Warning and Precautions

  • Serious gastrointestinal side effects may occur in patients treated with Ceritinib 750 mg under fasted conditions. Monitor and manage patients using standard of care, including antiemetics, antidiarrheals, or fluid replacement, as required. Withhold this therapy if a gastrointestinal side effect is critical or not tolerable and is not responsive to antidiarrheals or antiemetics.
  • Drug-induced hepatotoxicity may develop in patients treated with Ceritinib. Monitor with liver laboratory tests, including AST, ALT, and total bilirubin, monthly and as clinically required, with more periodic testing in those who develop transaminase elevations. Based on the severity of the side effects, withhold treatment with resumption at a decreased dose, or permanently stop treatment.
  • Severe, critical, or fatal interstitial lung diseases (ILD)/pneumonitis may occur in patients treated with Ceritinib capsules. Monitor patients for ILD/pneumonitis. Exclude other probable causes of ILD or pneumonitis and permanently stop treatment in those diagnosed with treatment-related ILD or pneumonitis.
  • Hyperglycemia may be established in patients treated with Zykadia 150 mg capsules. Monitor fasting serum glucose before the start of the treatment and periodically thereafter as clinically needed. Start or optimize anti-hyperglycemic medicines as required.
  • Bradycardia may occur in patients treated with Ceritinib. Avoid using this drug in combination with other products known to cause bradycardia (such as beta-blockers, nondihydropyridine calcium channel blockers, digoxin, and clonidine) to the extent possible. Monitor heart rate as well as blood pressure on a regular basis.
  • Pancreatitis may develop in patients treated with Ceritinib 150 mg capsules. Monitor lipase and amylase before the start of treatment and periodically thereafter as clinically required. Based on the seriousness of the laboratory abnormalities, withhold therapy with resumption at a reduced dose.
