Afatinib Tablet, a tyrosine kinase inhibitor (TKI), targeted treatment that irreversibly inhibits the ErbB family of tyrosine kinases. It is approved as a first-line therapy to treat locally advanced or metastatic non-small cell lung cancer (NSCLC) that harbors nonresistant epidermal growth factor receptor (EGFR) mutations, and as a second-line therapy to treat advanced squamous non-small cell lung cancer (NSCLC).
The recommended dosage of Afatinib is 40 mg administered orally once daily until disease progression or no longer tolerated by the patient. Take this therapeutic drug at least 1 hour prior to or 2 hours after food/meal. Do not take any missed dose within 12 hours of the next scheduled dose. Â
The most common side effects of Afatinib are nausea, vomiting, diarrhea, rash/acneiform dermatitis, dry skin, stomatitis, paronychia, decreased appetite, and pruritus.
Diarrhea may result in dehydration and renal failure. Withhold Afatinib tablets for severe and prolonged diarrhea not responsive to antidiarrheal agents. Provide an anti-diarrheal agent (e.g., loperamide) for self-administration at the onset of diarrhea and instruct patients to continue anti-diarrheal treatment until loose bowel movements cease for 12 hours.Â
Interstitial lung disease or ILD-like side effects (such as lung infiltration, acute respiratory distress syndrome, pneumonitis, or alveolitis allergic) may occur. Withhold Afanat during the evaluation of patients with suspected interstitial lung disease and stop therapy in those with confirmed ILD.
Obtain periodic liver testing in individuals during therapy with Afanat tablets. Withhold therapy in patients who develop worsening of liver function. In individuals who develop severe hepatic impairment while taking this medicine, discontinue treatment.Â
Keratitis, characterized as acute/worsening eye inflammation, light sensitivity, blurred vision, eye pain, lacrimation, and/or red eye may occur in patients treated with Afatinib. This drug should be used with caution in those with a history of keratitis, ulcerative keratitis, or serious dry eye.
