Description

Ribociclib, a kinase inhibitor, is prescribed to treat adult patients with HR-positive, HER2-negative advanced or metastatic breast cancer when combined with:

  • An aromatase inhibitor as initial endocrine-based therapy; or
  • Fulvestrant as initial endocrine-based therapy or following disease progression on endocrine therapy in postmenopausal women or in men.

Dosage and Side Effects

The recommended dose of Ribociclib is 600 mg, which is three 200 mg tablets. Take them by mouth once a day for 21 days straight, then take a break from treatment for 7 days. This makes a full cycle of treatment last for 28 days. Ribociclib tablets are to be taken by mouth, with or without food, alongside an aromatase inhibitor or fulvestrant. Swallow tablets whole; do not chew, crush, or split them. Do not ingest any tablet that is broken or damaged.

  • Severe skin reactions, such as Stevens-Johnson Syndrome (SJS), toxic epidermal necrolysis (TEN), and drug-induced hypersensitivity syndrome (DiHS)/drug reaction with eosinophilia and systemic symptoms (DRESS), may develop in patients using Ribociclib. If any signs or symptoms of these reactions appear, pause treatment until the cause is identified. If SJS, TEN, or DiHS/DRESS is diagnosed, discontinue Ribociclib permanently.
  • Elevations in serum transaminase levels can happen. Before beginning treatment with Ribociclib 200 mg, conduct liver function tests (LFTs). Monitor LFTs every two weeks for the initial two cycles, at the onset of each subsequent four cycles, and as deemed necessary based on clinical judgment.
  • Ribociclib can be responsible for causing fetal harm when administered to a pregnant woman. Women of reproductive potential should use effective contraception during treatment with Ribociclib and for at least 3 weeks after the final dose.
