Description

Carboplatin is an intravenously administered platinum coordination complex and alkylating agent that is used as a chemotherapeutic agent for the treatment of advanced ovarian carcinoma but is also used in other solid tumors including lung, and head and neck cancer. 

Dosage and Side Effects

Carboplatin as a single agent has been demonstrated to be effective in patients with recurrent ovarian carcinoma at a dosage of 360 mg/m2 intravenously (IV) on day 1 every 4 weeks. In the chemotherapy of advanced ovarian cancer, a promising combination for previously untreated patients consists of carboplatin injection 300 mg/m2 administered intravenously (IV) on day 1 every four weeks for 6 cycles, or cyclophosphamide 600 mg/m2 intravenously on day 1 every four weeks for 6 cycles.

The most common side effects of Carboplatin are nausea and vomiting (may be severe), loss of appetite, stomach pain, feeling tired, temporary hair loss, body aches or pains, diarrhea, constipation, weakness, and injection site reactions (redness, pain, and swelling).

Warning and Precautions

  • Bone marrow suppression (neutropenia, leukopenia, and thrombocytopenia) is dose-dependent and is also the dose-limiting toxicity. Peripheral blood counts need to be frequently monitored during Carboplatin 150mg/15mL or 450mg/45mL treatment and, when suitable, until recovery is achieved.
  • Carboplatin treatment may be responsible for causing fetal harm if administered to a pregnant woman. Females of childbearing potential/age should avoid becoming pregnant while receiving this medicine. 
  • Needles or intravenous (IV) use sets containing aluminum parts that may come in contact with carboplatin injection should not be used for the preparation or administration of the medicine. Aluminum may react with carboplatin causing precipitate formation and loss of potency.
  • Because there is a potential for toxicity in nursing infants secondary to Carboplatin 450mg/45mL or 150mg/15mL treatment of the mother, it is recommended that breastfeeding be stopped in case the mother is treated with carboplatin injection.
